HEMODAGRAM: PRINCIPLE OF OPERATION
A. RED BLOOD CELL AND PLATELET COUNT
The volume and the optical density of every cell individually are calculated. Accurate platelet counts in the clinically significant range (excluding microcytic red cells and cell fragments) minimize the number of manual counts.
ADVIA120 is the only analyzer with two dimensional platelet analysis which leads to safe results. Moreover the optical density of red blood cells is their hemoglobin content. Thus ADVIA120 has the ability to:
• Calculate the concentration of hemoglobin in every cell.
• Calculate the mean concentration of hemoglobin per red blood cell.
• Provide the distribution curve of the hemoglobin concentration per red blood cell.
• Calculate the distribution width of the hemoglobin concentration when it is a numerical indicator of anisochromia.
Β. HEMOGLOBIN COUNT
Additionally to what was briefly described above, hemoglobin is calculated simultaneously by the classic method
of cyan methemoglobin(or even with non-cyanide reagents), next the MCHand MCHC are calculated numerically.
If there is a deviation between the calculated values and those that are directly measured a sign from the analyzer appears. In this way we have the possibility to find possible problems in the calculation of the relevant (Hgb, Hct)
due to problems concerning the specific sample, for instance the examination of a lipemic or an icteric sample. These characteristics are unique for ADVIA 120 due to its advanced technology. Thus, the evaluation of the morphology of red blood cells in a quantitative way becomes reliable.
The criteria for classification are absolutely objective and are based not only on the volume distribution but also on the distribution of hemoglobin of the red blood cells.
C. CALCULATION OF WHITE BLOOD CELLS & LEUKOCYTE TYPE
After the cytochemical classification according to peroxidase activity a complete and definite differentiation between the types and the count of the white blood cells is accomplished. Simultaneously in a different reaction chamber, the number, the size and the lobes of the nucleuses are determined by LASER technology.
The determination of leukocytes by two different and worldwide accepted methods provides us the ability to identify different pathological conditions (nucleated red blood cells, blasts, immature cells). Moreover, it surpasses other analyzers because it clearly utilizes hematological parameters such as the nucleus morphology and the peroxidase content.It should be noted that peroxidase stain is a reference method for the identification of leukemia in the microscope.
D. RETICULOCYTES COUNT
ADVIA 120 provides unique diagnostic information about the size and the hemoglobin concentration of the reticulocytes.
According to the last parameter Chr, which is the only analyzer that provides it, we take important information e.g. for the response of patient with renal failure and patients under chemotherapy.
Please, collect whole blood in a tube containing EDTA as anticoagulant. If the samples are not going to be analyzed within 8 hours after the sampling, store them between 2 °C to 8 °C. After that it is the laboratory’s responsibility to transport the sample to its location.

For us every hemodiagram is a separate case of examination of a living tissue, such as the whole blood, in vitro. They are tested through internal and external quality controls that we have established for the reliability of the result and when needed, we provide you (with no extra cost) with information according to the microscopic examination of every sample.
